From Bangkok, at Mochit bus terminal, there are few buses going to Sangkhlaburi. The public bus company Transport Co LTD, offers two buses in the morning including one departing at 9.30 am. The trip takes 6-7 hours. Otherwise, you will have to go to Kanchanaburi. From there, local bus 8203 goes to Sangkhlaburi with a long stop in Thong Pha Phum. It leaves every 40 minutes and the price is 130 THB for a 4:30 drive. There are also minivans who regularly commute Kanchanaburi to Sangkhlaburi in about 3 hours.
Be careful Sangkhlaburi is very popular with Thai people on weekend and public holidays. If from Kanchanburi you will not have trouble finding a place in a the bus or in a minivan, from Bangkok, if possible, buy your ticket in advance.

To leave Sangkhlaburi, you have several options. You can take the Transport Co LTD air-conditioned bus to Bangkok which stop in Kanchanaburi with two departures per day, one in the early morning and one in late morning or the local bus to Kanchanaburiis. It slower and less comfortable, but a bit cheaper and there are more departures per day (including one to 1pm) or take a minivan to Kanachanaburi (or Thong Pha Phum) with the company that has its office at the end of the street with the 7 Eleven that runs along the market. Be careful minivans can be full very quick on weekends and public holidays. Note that to go to Thong Pha Phum, there apparently is also red songthaews that regularly depart from the market (one at 11am).

The train is the cheapest way to travel in Thailand especially in 3rd class. This is also where the contact with the population is the most interesting and incredible...
Taking the plance to travel in Thailand is often not that much more expensive than the bus especially with the numerous specials offers and it is so much faster !
